Lucille Layton is the mother of Hershel Layton, spouse of Roland Layton and grandmother of Alfendi Layton. She appears in flash backs when Professor Layton was still a student with husband, Roland, where they all resided in Stansbury. She also appears in Professor Layton and the Azran Legacy, currently living in London with her husband.

Lucille wears a pink sweater with a white collared shirt underneath, a blue skirt, a red apron, white tights and red shoes. She has white hair which is worn in a bun.

Lucille shows a loving nature towards her son and husband and often worries about their welfare. She always sends either Hershel Layton or Roland Layton out to look for one and another.

At a very young age, Hershel Layton and Jean Descole's parents, Leon and Rachel Bronev, were kidnapped by Targent due to their research on the Azran Civilization, leaving the two boys alone. The news of the orphans reached Lucille and her husband Roland, and so they decided to adopt Hershel, however Descole was originally planned to be adopted.

During Hershel's teens, they moved from London to Stansbury for three years, before moving back to London so that Hershel could study archaeology at Gressenheller University.

Chapter 2: The Secret of Norwell
After Hershel Layton returns home from school, he finds Lucille in a fit of worry. She tells him that some strange men in black came to see him, and Roland had taken them out and hadn't returned since. She asks Hershel to go out and search for him, while she stayed at home in case he returned. Upon their return, Roland reminds her that the people that were looking for him were his friends, and she tells him how worried she was and didn't know he would be out all day.

After Hershel spends time in his room, Lucille calls him down to dinner, and they discuss his day at school. She asks who would go to the next fencing tournament out of him and Randall, but Hershel tells her he wasn't about to let him win. She offers him pudding to strengthen him up, but he declines as he was about to go over to Randall's house, and she tells him not to stay out too late.

Hershel quickly drops in at home as he had forgotten something, so Lucille asks him to help her out with a puzzle while he was there, and afterwards lets him go.

Chapter 4: Angela's Tears
When Lucille sees that Hershel Layton brought Randall and Angela over, she offers them some cordial and biscuits, and noted she would have made some tea if she knew they would be coming over. Angela tells her not to worry, as they would be off again. She always tells them if she had more time, she would have given them some cake.

In a special episode, Hershel returns home from his trip to Randall's, and tells her and Roland he would go to bed early, to which she says was unusually sensible of him. After he leaves, Lucille asks Roland who the men were that he went out with, and he tells her it was a group of archaeologists who wanted to see the Wall of Norwell. He didn't know why they wanted to see Hershel, and after she expresses her worry, Roland suggests they both have some cocoa.

Chapter 6: The Ghost of Akbadain
Weeks after Randall's disappearance in the Akbadain Ruins, Hershel and his family decide to leave Stansbury, in order for him to study archaeology at Gressenheller University. Lucille asks if he was ready to go, but he wanted to take a moment to say goodbye to Randall. She told him that the carriage was ready when he was.

"A kind and spritely lady, Lucille is both a caring mother to Layton and an excellent chef, as the whole village can attest. She takes pride in keeping the house neat and tidy, and is all too used to collecting books, laundry and tea cups from under Hershel's bed."
